제목 Preparation of the PEG-crosslinked Chitosan Nanoparticles using Electrospinning for Delivery Carriers
초록 Crosslinked low molecular weight chitosan (LMC) using poly (ethylene glycol) (PEG) was synthesized with various LMC concentration and PEG molecular weights. The synthesis of PEG-crosslinked low molecular weight chitosan (P-CLMC) was confirmed by FT-IR, 1H-NMR spectra and DSC. Subsequently, ionically crosslinked electrospun nanoparticles based on P-CLMC were formulated with adenovirus DNA using the ionic gelation technique with tripolyphosphate anions as crosslinking agent. We were observed the TEM and measured the DLS to investigate the morphology and size of the P-CLMC nanoparticles. The resulting P-CLMC nanoparticles including the adenovirus DNA were investigated with regard to in vitro transfection efficiency and cellular uptake using fluorescence microscopy and flow cytometry and toxicity with MTT assay using ELISA. Thus, the prepared P-CLMC nanoparticles could be a better delivery carrier than chitosan in the development of delivery carriers
