Synthesis and Characterization of Anisotropic Polypeptide Nanoparticles with Opposite Charges for Advanced Drug Delivery |

Biopolymer-based anisotropic nanostructures with distinct multi-compartments could be of great interest for development of advanced drug delivery systems because they are capable of controlling drug release kinetics at each compartment. In this study, anisotropic elastin-based polypeptide (EBP) nanoparticles with discrete polarity were synthesized and characterized. Two oppositely charged EBPs were genetically engineered, expressed in E. coli and purified via inverse transition cycling, followed by introduction of hydrophobic moieties to the functional groups of EBP side chain for stabilization. Furthermore, dipolar EBP nanoparticles with oppositely charged compartments were prepared with two different hydrophobically-modified EBPs via electrohydrodynamic co-jetting and their colloidal assembly was evaluated. Potentially, the anisotropic polypeptide nanoparticles with opposite charges could be useful for drug delivery, molecular imaging and guided self-assembly. |
