제목 Molecular Design, Expression and Characterization of Elastin-Resilin based Block Copolymers for Injectable Hydrogels
초록 Protein-based block copolymers for injectable hydrogels have been widely studied for functional tissue engineering applications. In this study, elastin-resilin based block copolymers with different block lengths were genetically engineered, expressed, and purified by inverse transition cycling. This work was motivated to combine unique thermo-responsiveness of elastin-based polypeptides (EBPs) with high resilient property of resilin-like polypeptides (RLPs) for formation of self-assembled hydrogel network with high elasticity and resilience. The EBP blocks at both ends formed hydrophobic aggregate domains above their transition temperatures while the RLP blocks with high resilience being hydrated, resulting in formation of physically cross-linked hydrogels via temperature-triggered self-assembly. In conclusion, these elastin-resilin based block copolymers would be promising as injectable hydrogels for tissue engineering and regenerative medicine.
