Synthesis of lipoic acid-peptide conjugates and their effect on collagen and melanogenesis |

In this study PEGylated lipoic acid (LA-PEG) was conjugated to some known anti-wrinkle oligopepetide structures. The yield was 42% after 12h reaction time. The physiological activities such as cytotoxicity, whitening effect, and anti-wrinkle effect of the target PEGylated peptides synthesized lipoic penta (LA-PEG-KTTKS) were investigated. A tyrosinase inhibition analysis showed the inhibitory effect up to 78.4% at the concentration of 0.2mM and the melanin generation inhibition assays were reduced to 56.3% at 0.2 mM. Wrinkle effect was investigated through MMP-1 inhibitory activity assay and collagen synthesis ability. MMP-1 inhibitory activity was consistently inhibitory in the entire concentration range of 0.01 to 0.5 mM. At over 0.25mM, MMP-1 inhibitory activity of LA-PEG-KTTKS exhibited 30% higher effective than that of LA-PEG. lipoic acid, the amount of collagen was found to be increased about 40%. |
