Preparation of Surfactant-free Nanoparticles of Methotrexate-conjugated Poly(DL-lactide-co-glycolide) |

In this study, we have synthesized methotrexate (MTX)-conjugated poly(DL-lactide-co-glycolide) (PLGA) and prepared nanoparticles without use of surfactant. Before MTX conjugated to the end PLGA chain, hexamethylenediamine (HMDA) was conjugated to the carboxylic end of PLGA. After that, MTX was conjugated to end of amine group of PLGA-HMDA conjugates. Chemical composition of MTX-conjugated PLGA was confirmed with 1HNMR, indicating that MTX has intrinsic peaks between 5 and 8 ppm while PLGA showed specific peaks between 1 and 6 ppm. Nanoparticles was prepared by nanoprecipitation and dialysis method. Morphological observation with TEM showed spherical shapes with particle size lower than 500 nm. |
