C2C12 myotubes on the application of electrical signals lead the development of muscle tissue engineering including bio-actuators and muscle tissue regeneration. To maximize the contractility of electrical signals, an alignment of C2C12 myotubes is required. In this study, we present a new strategy to enhance myogenesis in C2C12 skeletal muscle cells via magnetism of metallic nanoparticles. We used functionalized iron oxide nanoparticles (IONPs) with amine or NHS ester groups to conjugate with biomaterials such as fibronectin and RGD sequence to improve cellular uptake. These conjugated IONPs were facilitated not only cellular uptake, but unidirectionally cultivate the myotubes in vitro. The formation of myotubes was controlled by the magnetic field varying strength, period, and interval. Overall, this new strategy to align C2C12 myotubes using biomaterials and functionalized IONPs would be actively utilized for cell culture system and bio-robots in the tissue engineering. |
