Polyethylenimine (PEI) has been used efficient gene carrier because of their cationic property and DNA condensing ability. PEI with high molecular weight has high gene delivery efficiency, but their toxicity is high, too. Polyamidoamine (PAMAM) dendrimer is known for efficient carrier because of their endosome buffering effect and biodegradable and safe, nonimmunogenic properties. In this study, we synthesized low molecular weight PEI based PAMAM as a vector for gene delivery. And, L-Arginine (R) is conjugated primary amine group of this novel polymer surface to generate R-grafted PEI-PAMAM. Also, we synthesized dexamethasone-grafted PEI-PAMAM. This polymer can mediate nucleus-targeting gene delivery. As a result, these novel polymers showed the outstanding gene delivery efficiency and relatively low toxicity in comparison with other gene carriers. The study indicates that these novel polymers possess potential to be a promising dendritic gene carrier for gene therapy. |
