제목 Polymorphic Selection of Metastable Form by Controlling Drowning-out Crystallization
초록 Pharmaceutical development of thermodynamically metastable forms is often desired because of their enhanced biopharmaceutical properties as a result of higher solubilities and faster dissolution rates.
In this study, metastable form of an active pharmaceutical ingredient (API) was achieved by controlling drowning-out crystallization conducted using acetone as an antisolvent in aqueous solution with a batch crystallization mode. Some techniques were manipulated to monitor in-line the phase transition of crystallization such as the focus beam reflectance method (FBRM) and particle vision measurement (PVM). The polymorphs of API was identified by Raman spectroscopy and confirmed by XPRD. In higher supersaturation conditions, there is a trend to form metastable polymorph whilst in lower supersaturation conditions the selection of stable polymorph often occurs in this drowning-out crystallization process.
