Improved expression of cell penetrating scFv using an engineered ubiquitin in a cell-free protein synthesis system |
초록 |
Antibody is in the core of current research for novel therapeutic and diagnostic agents. In particular, antibodies that can cross cellular membranes have potential applications in controlling disease-related intracellular protein-protein interactions. In this study, we developed an engineered ubiquitin sequence that substantially improves the expression level of cytosol-penetrating antibodies in a cell-free protein synthesis system. The initial codons of a ubiquitin gene was randomized, and the expression level of scFv fused with each of the variant ubiquitin sequences was examined to identify the optimal initial codons. As a result, we were able to select a number of variant ubiquitin clones that significantly improves the expression level of the fused cytosol-penetrating antibodies. |
