Engineering thermal property of elastin-like proteins (ELPs) through incorporation of unnatural amino acids in a cell-free protein synthesis system |
초록 |
Incorporation of unnatural amino acids into proteins is of particular interest in that it can endow proteins with novel properties that cannot be met with the natural set of 20 amino acids. We adopted and modified a cell-free protein synthesis system to incorporate unnatural amino acids into a variant elastin- like protein. Through the residue-specific substitution of proline with a number of steric analogues, we were able to change the temperature-responding characteristic of the polymer depending on the kinds of substituting amino acids. This study demonstrates the possibility of using cell-free protein synthesis systems for producing engineered bio-materials with expanded amino acid repertoire. |
