Targeting of green fluorescent proteins into Lysosomes using DAP2 signal peptide |

Most proteins consist one or more long peptide chains with signal peptide. Those exist different positions in the proteins and have various mechanisms because of signal peptide. Notably, One of the signal peptide on the lysosomal protein is DAP2 which is on the Dipeptidyl aminopeptidase B(DPAP B). DAP2 signal peptide function is to transport the protein from the Golgi to the lysosome. In our study, we developed of transfer vector with DAP2 signal peptide to transport green fluorescent proteins(GFP) to the lysosome. The expression of GFP in recombinant yeast was observed using fluorescence microscope. Interestingly DAP2 signal peptide is successfully help to delivery GFP to the lysosome. This is designed for delivery any protein which don’t exist in lysosome. It would be useful backbone vector to delivery any proteins to yeast lysosome as molecular tool in eukaryotic cells.
