Development of Disulfide Stabilized Heterfunctional Pluronic micelle for Effective Intracellular Anti Cancer Drug Delivery |

Heterfunctional Pluronic-SH (Plu-SH) nano particles have been developed as articulate target specific anticancer drug carrier for intracellular drug release triggered by glutathione in tumor cells. The core of the micelles formed disulfide bond when dissolved in aqueous solution and encapsulated hydrophobic drug molecules within its hydrophobic core with effective stability at physiological pH. The size of these unique nanoparticles was around 95.6nm determined by the dynamic light scattering. But in reducing environment the polymeric micelles disassembled due to the cleavage of disulfide bond and the drug loading micelles showed long time drug release in presence of 70mM Dithiothreitol (DTT). The paclitaxel loaded micelles showed around 38% cell viability in A549 cells that suggests effective cytotoxicity can be achieved through these nanoparticles improving drug delivery system. |
