A hybrid nanogel of L3 phased porous silica and polymer has been developed based on interpenetrating network of thermo-responsive poly(N-isopropylacrylamide) gels and tailored nanoporous silica. A sustainable positive thermo-responsive release profile on the anticancer drugs is obtained. When the temperature rises, the polymer gel shrinks, squeezing the drug into the porous channels, and at the same time, opening the pore to the outside media. The drug slowly diffuses out of the porous channels. The overall release rate can be adjusted by changing the composition of the nanogel. The dug release mechanism is proposed to achieve a remarkable positive thermal-responsive on-off regulation for indomethacin release from hybrid nanogels. The mechanism allows an uniform release rate when the temperature is maintained above the LCST. In addition, changing composition of the nanogel ( the water content ) significantly extends the effective release over a long period of time. |
