Metabolic Engineering of Corynebacterium glutamicum for Production of 1,5-diaminopentane |
초록 |
The bio-based production of 1,5-diaminopentane (cadaverine) from renewable feedstock is a sustainable and promising alternative to petroleum-based chemical synthesis. A metabolically engineered Corynebacterium glutamicum to produce 1,5-diaminopentane is reported here. L-lysine decarboxylase was amplified by plasmid-based overexpression of the cadA gene under the tac promoter in an industrial L-lysine producer C. glutamicum (U2 strain). With the redesigned cadA gene, a production titer of 31.94 g/L of 1,5-diaminopentane was achieved without exogenous feeding of L-lysine by fed-batch fermentation of C. glutamicum (U2/pCEcadA).
