We investigated the effect of amphiphilic comb polymer on reducing nonspecific adsorption of biomolecules in membrane filtration. The amphiphilic comb polymer is MMA-based coating material with a short nonfouling oligoethylene glycol side-chain that enables long-term, bio-repellant surfaces. The MMA based nonfouling polymer was synthesized by free radical polymerization of methylmethacrylate (MMA), poly(ethylene glycol) methacrylate and poly(ethylene glycol) methyl ether methacrylate. The number average molecular weight (MW) of the comb polymer (Mn) was ~30,000 Da with a polydispersity of ~2.3, as measured by gel permeation chromatography using PS calibration standards. The composition of the terpolymer was characterized by 1H NMR in CDCl3. The nonfouling property of the polymer on a commercially available filtration membrane was observed with the adsorption experiment of cell-lysate solutions. In the paper, we suggested feasible evaluation methods to quantify fouling phenomena by adopting biological tools. |
