Preparation of Methotrexate incorporated polymeric micelles of methoxy poly(ethylene glycol)-grafted chitosan. |
| 초록 |
We synthesized methoxy poly(ethylene glycol) (MPEG)-grafted chitosan (ChitoPEG) copolymer to prepare methotrexate (MTX)-encapsulated polymeric micelles. MTX-encapsulated polymeric micelle of ChitoPEG copolymer was prepared by mixing of MTX/DMSO solution into ChitoPEG/deionized water. It was suggesyted that polymeric micelles are self-associated through ionic complex formation between chitosan main chain and MTX. MTX-encapsulated polymeric micelles showed spherical shapes and their particle size was around 50 nm at TEM observations. To approve core-shell structure of self-associates, 1H NMR study was performed. When polymeric micelle was dissolved into DMSO or DCl/D2O mixtures, the specific peaks of MTX and ChitoPEG was observed while their specific peaks were disappeared at D2O and peaks of PEG was observed, indicating that self-associates between MTX and ChitoPEG copolymer formed micellar structure. |
