As previous studies, polyamidoamine(PAMAM) dendrimer is widely known as a useful polymer for drug and gene delivery system. PAMAM dendrimer has primary, secondary and tertiary amine groups which can make complexes with plasmid DNA. and show good transfection efficiency and low cytotoxicity. And PAMAM G4 has 64 terminal amine groups which could be modified with biological or chemical molecules. In this study, we conjugated two amino acids at terminal groups of PAMAM G4. We combined arginines and prolines on terminal amino groups of PAMAM G4 with HOBt and HBTU coupling reagent. In addition, we conducted the experiments of complex formation assay with gel electrophoresis and picogreen assay test. Then, we performed the transfection efficiency assay and cytotoxicity test for NIH 3T3 and HeLa cell lines. As a result, these surface modified PAMAM G4 showed significant gene transfection efficiency and low cytotoxicity. |
