To develop polymeric drug delivery carriers for cancer therapy, arginine-grafted poly(amido ethyleneimine)/heparin complex was formulated. Poly(amido ethyleneimine) was polymerized by Michael addition between N,N’-cystaminebisacrylamide (CBA) and N-Boc-1,6-diaminohexane (DAH). Arginine was conjugated to the primary amine group of poly(CBA-DAH) to increase the cellular uptake. Arginine-grafted poly(CBA-DAH) (AGP) which has disulfide bond could be cleaved by intracellular reducing agents such as glutathione in cytoplasm. Cationic AGP and anionic heparin were formed nano-sized particles by the electrostatic interactions. As a cancer drug, vinblastine was loaded into particles by physical loading. AGP/heparin/vinblastine complex was the proper size (less than 200nm) and it was positively charged. Poly(CBA-DAH) grafted with arginine is successfully developed for drug delivery carriers in to the cytoplasm. |
