제목 Spurred Gene Transfer through Stimulating Osmotic Fragility of Endosome by Polyxylitol-based Transporter
초록 Endosomal escape is one of the most important processes in the efficient non-viral gene delivery system and far much efforts have been taken to enhance this step. Here, we designed the new non-viral vector, polyxylitol-based transporter(PXT) having enhanced endosomal escape through stimulating osmotic fragility of endosome. PXT was synthesized through Michael addition reaction between xylitol diacrylate as a crosslinker and low molecular weight PEI(1.2K) and it showed higher cell viability and transfection efficiency than those of PEI 25kDa. The small amount of xylitol integrated in PXT(4% w/w) made one-third of osmotic pressure in PXT and this made endosome more swell and fragile, thus yielding pDNA at least 10-fold more released into cytosol and 4-fold more localized to nucleus compared with PEI. Probably this mechanism will be also valid to post-mitotic, differentiated C2C12 cells and we expect that the PXT might be a efficient gene carrier through this endosome-escaping mechanism.
