Building In-Vitro Neuronal Network Patterns and Controlling Its Functions via Vertical Nanowire Arrays |

In order to investigate the relation between in vitro neuronal network function and its connectivity formation and modulation among the network’s constituent neurons, it is a critical step to guide the axonal growth and control the initial connectivity in a given network. Here we present a new means of building in vitro neuronal networks using vertical nanowires. Primary culture of rat hippocampal neurons were deposited on patterned nanowire arrays and maintained for several weeks. We observed that neuronal networks are formed along the patterned nanowire lines while their viability remains up to 5 weeks even without cell adhesive coatings on the substrate. Combined with the recently-developed transfection method via nanowire internalization, the patterned vertical nanostructures will contribute to understanding how synaptic connectivity and site-specific perturbation will affect global neuronal network function in an extant in vitro neuronal circuit. |
