제목 | Synthesis of cell penetrating peptide conjugated nanoparticle for hearing loss |
초록 | Hair cells, which are located in the cochlear of the inner ear. Once hair cells are damaged, they cannot grow and cause permanent hearing loss. The therapies for hearing loss with traditional strategies were often unsuccessful, because only small quantities of drug could access to hair cells through cochlear. In this study, we developed drug delivery system for hearing loss. We synthesized nanoparticles which can access to hair cell through round window membrane (RWM). These are polymer micelles, so hydrophobic drug located in the inside of the shell, and poly(ethylene glycol) is decorated on the outside. In addition, we used the cell penetrating peptide to the nanoparticle to penetrate RWM and hair cell membrane. The structures of molecules were analyzed using NMR. The surface charge of nanoparticles was positive, and the size was less than 50nm. These results suggest that the CPP conjugated nanoparticles can be drug carrier for hair cell, and we will conduct In vivo and In vitro experiment to confirm this. |
