Co-expression of fluorescence protein for assessment of gene expression in a cell-free protein synthesis system |

The assay of recombinant protein expression generally requires time- and labor-intensive steps. In this study, we demonstrate the use of a cell-free protein synthesis system as a tool for assessment of translational efficiency of different genes. We estimate that the signal intensity from the reporter protein inversely represented the expression level of the co-expressed proteins during the co-expression of a gene of interest with a reporter gene. This allowed us to measure the relative translational efficiencies of different genes in a high-throughput manner. Furthermore, relative translation efficiencies determined by the cell-free co-expression experiments were successfully reproduces when the same genes were deploy into Escherichia coli, suggesting that this method can also be used to predict the yield of protein production a cell-based gene expression. |
