Delivery of siRNA with Polymeric Nanoparticles for Cancer Therapy |

Delivery of siRNA remains the major obstacle to therapeutic application of RNA interference. The biodegradable amphiphilic and cationic triblock copolymer self-assembles into micellar nanoparticles in aqueous solution, which further form micelleplexes with siRNA. A micelleplex carrying siRNA targeting an oncogene induces remarkable apoptosis and reduces the proliferation of cancer cells, leading to significant inhibition to tumor growth following systemic administration. It is further demonstrated that the micelleplex can simultaneously deliver paclitaxel and PLk1-siRNA to the same tumoral cells both in vitro and in vivo following i.v. injection, which leads to a synergistic tumor suppression effect in the tumor xenograft murine model, requiring a thousand-fold less paclitaxel than needed for paclitaxel monotherapy delivered by the micelleplex and without activation of the innate immune response or generation of carrier-associated toxicity. |
