Functional expression of oleandomycin glycosyltransferase gene from Streptomyces antibioticus and its application to glycosylation |
초록 |
Glycosylation is a glycosyl-transfer reaction to enhance biological activities of secondary metabolites such as flavonoids, alkaloids and terpenoids. Glycosyltransferases cancatalyze the transfer of a sugar moiety from an activated nucleotide sugar to an acceptor molecule. The oleandomycin glycosyltransferase (OleD) gene from Streptomyces antibioticus was functionally expressed in Escherichia coli BL21 (DE3) in the presence of various molecular chaperones. The functional recombinant OleD with a his tag was purified using a His-tag affinity column, and tested for its ability to glycosylation of secondary metabolites with uridinediphosphate-activated glucose. |
