Surface Immobilization of Anti-Thrombin Aptamers on Si-Nanowire for Biosensor Applications |

In this work, aptamer biosensor (aptasensor) gate was prepared to detect the optical and electronic signals during and after the chemical binding of biomolecules to aptamers on the chip. A silicon-nanowire (Si-NW) was used as a model substrate and anti-thrombin aptamer was used as a model aptamer. Surface modification was carried out using 3-aminopropyl diethoxysilane and succinic anhydride, or 3-trimethylsilylpropyl methacrylate to provide Si-NW with carboxyl or methacrylate groups. Aptamers with amine or thiol groups on 5’-end were chemically immobilized on the surface-modified Si-NW. Atomic force microscopic analysis confirmed the successful binding of anti-thrombin aptamers on Si-NW. The binding capacity of the aptamer biosensor to FITC labeled thrombin was successfully assessed with a fluorescent microscope. The thrombin aptasensor gate will be further applied for the real-time detection of electronic signals during and after thrombin binding. |
