제목 Probing the dynamic profiles of cytokine secretion from individual human CD4+ T cells responding to antigen
초록 In this study, we use a single-cell co-culture assay to temporally resolve antigen-dependent cytokine responses exhibited by human CD4+ T cells ex vivo. Monocyte-derived human dendritic cells pulsed with peptides from common antigens (influenza, tetanus toxoid, cytomegalovirus, and Epstein-Barr virus) were co-incubated in arrays of microwells with individual autologous human CD4+ T cells, and the secretory responses were measured for three different analytes at two time points.  The results show that the reported single cell assay can probe the profile of functional responses from individual antigen-reactive T cells persistently stimulated by antigen-presenting cells.
