제목 | Effective bone tissue regeneration using 3D printed PCL scaffold incorporating ECM enriched hydrogel |
초록 | Recently, synthetic polymers often used for 3D printing lack the biological functionality to mimic native extracellular matrix effects to support mesenchymal stem cells (MSCs) growth. In this work, we describe the fabrication and characterization of a 3D model for tissue engineering that comprises an extracellular matrix (ECM)-enriched hydrogel incorporated into a polycaprolactone (PCL) scaffold. The matrisome (MTS) was utilized as a source of ECM-enriched biomacromolecules, and biocompatibility was evaluated in vitro using human bone marrow-derived MSCs (hMSCs). The 3D complex model demonstrated good viability and osteogenic differentiation of hMSCs in vitro in both two-dimensional (2D) and 3D cultures. The complex was also evaluated in vivo using a rat calvarial defect model. Therefore, we suggested that MTS-based 3D complex model may serve as an effective tissue scaffold for use in the field of tissue engineering. |
