The evaluation of biocompatibility of Epoxy and PDMS in vitro as a implantable coating materials |

Epoxy and polydimethylsiloxane (PDMS) are industrially important polymers used for the implantable coating material, according to their certified biocompatibility, sufficient mechanical strength and easy processibility. The cytotoxicity of these materials was evaluated using the MTT[tetrazolium dye, 2-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide, C18H16N5SBr] assay. Materials extract was prepared following ISO 10993-5 procedure for preparing extracts of materials. We chose NIH-3T3 fibroblast cells because they represent well-characterized and established cell cultures for cytotoxicity assays. Positive controls were prepared by extracts from organo-tin based PVC, SDS solution and Triton-X 100 solution. Two different methods were adopted for the MTT test: 1) seeding cells with extracts 2) incubation extracts with cell sheets. In the presentation, both cell morphology and MTT numberal data will be shown for the comparison. |
