In this study, adenovirus encapsulated alginate beads were fabricated successfully by electrospinning method. Bead morphology and size were characterized by optical microscopy. As the concentration of alginate increased, bead size increased slightly. Crosslinking density of alginate beads increased with an increase of the concentration of calcium chloride. Encapsulation degree of alginate beads was characterized by confocal laser microscopy. The virus encapsulated beads were loaded onto the cancer cells and then green fluorescence protein (GFP) expression levels were measured by the fluorescence intensity analysis. Adenovirus encapsulated alginate microbeads resulted in sustained release of virus with slow rate for a long period due to biodegradable alginate shell, which can maintain the treatment effects. This result was corroborated by FACS analysis. This method may be useful for delivering viral vectors in other tissues. |
