To develop chitosan based gene carrier, chitosan oligosaccharide (COS) were chemically modified with hydrophobic bile acids (BAs) with different hydrophobicities. The physicochemical properties of the hydrophobized COSs (COSBAs) were investigated by using dynamic light scattering. The COSBAs formed nano-sized particles in aqueous milieu by hydrophobic interaction between the BA groups. Than COSBAs were complexed with DNA, and COSBA/DNA complexes were characterized. The COSBAs efficiently condensed the plasmid DNA, and protected the condensed DNA from endonuclease attacks. Furthermore, some COSBA/DNA complexes showed dramatically enhanced gene transfection efficiencies than unmodified COS and control group of PLL with negligible cytotoxic effects. Therefore, the COSBAs have much potential for efficient gene carrier. |
