Synthesis and characterization of Dextran-Spermine-gCB polymer as a efficient targeted gene carrier. |
초록 |
Dextran, a biodegradable natural polysaccharide has been modified with cationic oligoamine, spermine as a non-viral polymeric gene carrier. In this study, D-spm which is a cationic polysaccharide based on spermine-dextran was successfully synthesized by chemical modification after reductive amination. In addition, glactosyl conjugated cucurbituril(galCB) was bound into the cationic polysacchardes via host-guest interaction of D-spm and galCB bridge to exhibit higher cell specific-targeting efficiency simultaneously. The formation and substitution degree of oligoamines on D-spm/galCB were confirmed by NMR, IR and elemental analysis. The physicochemical properties of D-spm/galCB were evaluated by particle size measurement and zeta potential assay. Also, the transfection efficiency and cytototixity of modified dextran were investigated in HepG2 cell containing hepatocyte receptor and HeLa cell, not containing it. |
