제목 Photosensitizer-conjugated Magnetic Iron Oxide/Gold (Core/Shell) Nanoparticle as a Platform for Controllable Photodynamic Cancer Therapy
초록 A core/shell structured magnetic iron oxide/gold (Fe3O4/Au) nanoparticle (NP) is investigated as a novel photosensitizer carrier for photodynamic therapy (PDT). A thiolated heparin-pheophorbide a conjugate (H-PhA) is synthesized and introduced onto the NP surface through gold-thiol interaction. Fe3O4/Au NP can act as a quenchable nanocarrier for H-PhA conjugate, whereas glutathione can mediate the H-PhA release in a photoactive form. In vitro and in vivo studies show that Fe3O4/Au/H-PhA NPs are effective for PDT due to the prolonged circulation characteristic, enhanced tumor specificity, and the controlled photoactivity. In addition, in vitro MRI studies reveal that the NPs could potentially serve as MRI contrast agent for cancer diagnosis, and monitor photodynamic treatment response for guiding accurate light irradiation. The present findings show that the Fe3O4/Au/H-PhA NP is a promising nanomedicine for cancer theranosis.
