Preparation and controlled photoactivity of photosensitizer conjugated heparin-gold nanoparticle for photodynamic therapy |

In this study, a novel pheophorbide a-conjugated hybrid nanoparticle with controllable photoactivity was design and synthesized. Pheophorbide a-conjugated heparin (PhA-H) was synthesized to encapsulate AuNP through gold-thiol interaction. Due to the strong fluorescence quenching effect of AuNP, the photoactivity of PhA-H/AuNP could be effectively prevented in physiological conditions. However, the photoactivity of PhA-H/AuNP could be restored by GSH-mediated intracellular dissociation of PhA-H molecules from AuNP. In vitro cell studies revealed that the PhA-H/AuNP could release the PhA-H molecules in photoactive form after cellular uptake and could generate singlet oxygen with light irradiation. Furthermore, PhA-H/AuNP presented prolonged blood circulation, enhanced tumor specificity, and improved therapeutic efficacy compared to free PhA as studied on tumor-bearing mice. These finding indicated PhA-H/AuNP could be used as safe and smart nanomedicine for clinical PDT. |
