Cloning and expression of ncs52 and ncs53 from neocarzinostatin Streptomyces carzinostaticus ATCC 15944 |
| 초록 |
The ncs52 and ncs53 are located in the neocarzinostatin biosynthetic gene cluster isolated from Streptomyces carzinostaticus. The ncs52 gene (765bp) encoding for the putative mannose-1-phosphate guanyltransferase, containing 255 amino acids with a calculated molecular mass of 28.05 kDa while the ncs53 gene (1kb) encoding for the putative UDP-glucose-4-epimerase, containing 333 amino acids with a calculated molecular mass 36.63 kDa. For the elucidation of putative function of ncs52 and ncs53 genes, expression plasmids were made by cloning in pET-32a(+) and overexpressed in E. coli BL21 (DE3) expression host with His-tag fusion protein at N-terminal. |
