제목 Cloning and expression of ncs52 and ncs53 from neocarzinostatin Streptomyces carzinostaticus ATCC 15944
초록 The ncs52 and ncs53 are located in the neocarzinostatin biosynthetic gene cluster isolated from Streptomyces carzinostaticus. The ncs52 gene (765bp) encoding for the putative mannose-1-phosphate guanyltransferase, containing 255 amino acids with a calculated molecular mass of 28.05 kDa while the ncs53 gene (1kb) encoding for the putative UDP-glucose-4-epimerase, containing 333 amino acids with a calculated molecular mass 36.63 kDa. For the elucidation of putative function of ncs52 and ncs53 genes, expression plasmids were made by cloning in pET-32a(+) and overexpressed in E. coli BL21 (DE3) expression host with His-tag fusion protein at N-terminal.
