제목 | Tissue engineered spinal cord using bone marrow stromal stem cell seeded nerve guidance channeled PLGA scaffolds. |
초록 | Bone marrow stromal stem cells (BMSCs) are a source of stem-like and progenitor cells. BMSCs transplanted into the normal animal central nervous systems differentiate into neural cells. Transplantation of adult BMSCs directly into adult rat brain reduces functional deficits associated with stoke. Porous scaffolds have highly porous structure with good interconnection between pores for allowing cell adhension and growth1. Fabrication of tubular poly(L-lactide-co-glycolide) (75:25 by mole ratio of lactide to glycolide, PLGA) scaffold made by ice particle leaching method and investigated as potential were guidance channel. BMSCs were harvested from the femurs and tibias of adult female Fischer rat. BMSCs were suspended at 2×106 cell/scaffolds in a 20㎍/㎖BDNF solution and then PLGA scaffold was saturated with 10㎕ BDNF solution. Fischer rats received an implant consisting of the BMSCs suspended in the PLGA scaffold in the gap (T8~T9; 5 ㎜ and 3 ㎜) created by the spinal cord resection. For histochemical evaluation, the implants were removed after 4 and 8 weeks. Thin sections were cut from paraffin embedded tissue and histological section were stained H&E staining and immunohistochemical staining. Motor functional outcome measurements using the Basso-Beattie-Bresnehan (BBB) score2 perfomed weekly to 8 weeks post injury. It was observed that the effects of PLGA scaffold with BMSCs on neuroinduction are stronger than PLGA scaffold without BMSCs and blnak. Also, we observed that the motor functional outcome of 3 ㎜ defect model are more improve that 5 ㎜ defect model. Fig1.
