1,3-propanediol (1,3-PDO) is an industrially important chemical with increasing demand as an intermediate to fibers and resins. No natural microorganism can produce 1,3-PDO from glucose, which is the most popular carbohydrate and can be generated from lignocellulosic biomass. However, there are only very few scientific studies on the production of 1,3-PDO from glucose due to extensive patenting by DuPont and Genecor. Compared to recombinant E. coli strains developed by Dupont, Klebsiella pneumoniae has several advantages. It is natural producer of 1,3-PDO from glycerol and also coenzyme B12. Therefore, this study aims to empower our lab isolates K. pneumoniae J2B with capability to produce 1,3-PDO from glucose. The genes responsible for the production of glycerol from glucose were overexpressed and the oxidative glycerol assimilation pathways were modified. Furthermore Dha operon genes were overexpressed to increase the conversion rate of glycerol to 1,3-PDO. |
