Improved of 1,3-propanediol production from glucose by enhanced NAD(P)H regeneration through pentose phosphate pathway |

1,3-Propanediol (1,3-PDO) is an important platform chemical. Its production from glucose has been developed and commercialized by DuPont, but scientific details are not well disclosed. This study aims to develop an economical microbial cell factory using Klebsiella pneumoniae J2B. Especially, we focused on the improvement of NAD(P)H regeneration because 1,3-PDO production is often limited by the availability of this cofactor. The EMP pathway was modified to reduce carbon flux through glycolysis and the PP pathway was upregulated by overexpressing less NADPH-sensitive Zwf and Gnd instead of the native counterpart. One resulting strain could produce 1,3-PDO from glucose with the high yield of 0.36 g/g glucose. |
