제목 A biocompatible poly(ethylene glycol)-b-poly(DL-lactide) containing lysine units: synthesis, self-assembly, in vitro, and in vivo evaluation
초록 A biodegradable copolymer poly(ethylene glycol)-b-poly(DL-lactide)-LLys4 was prepared by acidolysis of polyethylene-b-poly(DL-lactide)-benzyloxycarbonyl-L-lysine (PEG-PLA-ZLL4) synthesized by the ring-opening polymerization (ROP) of benzoyloxycarbonyl-L-lysine N-carboxyanhydride (ZLys NCA) and amino-terminated PEG-PLA-NH2 as a macroinitiator. The polymeric micelles of PEG-PLA- LLys4 were cross-linked by reaction of the core lysine moieties with bis α,ω-NHS ester. Paclitaxel (PTX) as anti-cancer drug was encapsulated into polymeric micelles, and the in vitro release behavior were investigated for polymeric micelles. The in vitro cytotoxic activity of PTX-micelles in A549 was evaluated by MTT assay. The corresponding in vivo antitumor efficacy was assessed in nude mice bearing A549 tumor after intravenous administration. PTX loaded polymeric micelles exhibited antitumor activity and reduced toxicity on A549 cell in vivo.
