Synthesis of high fluorescent water soluble Poly(p-phenyleneethynylene) copolymer for DNA sensing |
초록 |
We designed and synthesized a new high fluorescent water soluble Poly(p-phenylene ethynylene)s for the DNA sensor using the conventional palladium-catalyzed Sonogashira–Hagihara copolymerization. Bifurcate ethylene glycol ((2-(2-(2-methoxyethoxy)ethoxy)ethoxy)propan-2-yloxy)groups and carboxylic acid groups at the end of side chains are introduced for giving a water solubility and as a DNA receptor. The PPEs showed excellent solubility in water but poor solubility in common organic solvents as a THF and chloroform. Amine functionalized complementary 15bp ssDNAs (5’-ACA TCC GTG ATG TGT-3’ and 5’-TGT AGG CAC TAC ACA-3’) are used for coupling with PPEs. We found the polymers coupled with ssDNA were completely soluble in 6XSSPE buffer solution and success of hybridization between the ssDNAs was confirmed with electrophoresis method. |
