Preparation and evaluation of density-controlled 3D culture using hexanoyl glycol chitosan hydrogel coated dishes |

Three-dimensional (3D) cell culture is known to improve cellular functions by creating physiologically more similar to in vivo environment than 2D cell culture. Our previous study has given a facile method for the efficient formation of 3D spheroids using non-adhesive N-acyl glycol chitosan coated dishes. Among them, hexanoyl glycol chitosan is highly effective and reproducible method for cell spheroids formation. However, HGC has limitations such as low gel stability for long-term spheroid culture. In this study, chemically crosslinked HGC hydrogels were introduced to improve long-term stability of coating layer. We synthesized UV-crosslinkable methacrylated HGCs (M-HGCs) and their hydrogels were characterized. Coating morphology and mechanical properties were observed by SEM analysis and rheological studies. We also confirmed the number of cells per spheroid by H&E staining and we figured out the possibility to control of the density of spheroids. |
