제목 | Chondrogenic differentiation of human bone marrow-derived mesenchymal stem cells and rat muscle-derived stem cells in 3D structures. |
초록 | Damaged articular cartilage is well known to have a limited potential for repair or regeneration due to avascularity and the presence of relatively few cells with low mitotic activity. Bone marrow derived mesenchymal stem cells (BMSCs) and muscle derived stem cells (MDSCs) may represent a convenient source of stem cells for cell-mediate gene therapy and tissue engineering application. Also, they have both self-renewal capabilities and multipotentiality. In this study, we have demonstrated the chondrogenic potential of human BMSCs and rat MDSCs under 3D cell culture conditions, provided that growth factors and other beneficial ingredients are present. In vitro, BMSCs and MDSCs were seeded in polymeric scaffolds for 28 days. The chondrogenesis of BMSCs and MDSCs was characterized by RT-PCR, ALP, MTT assay and staining with hematoxylin-eosin, safranin-O. We have confirmed expression of cartilage specific marker genes such as type Ⅱ collagen and aggrecan by RT-PCR.
