제목 | Preliminary study on the adhesion and proliferation of bone marrow stem cells on demineralized bone particle impregnated PLGA scaffold. |
초록 | Current tissue engineering techniques require the use of a porous biodegradable/bioresorbable scaffold, which serves as three-dimensional template for initial cell attachment and subsequent tissue formation, both in vitro and in vivo. Natural biomaterial was demineralized bone particle (DBP). It seems that local prolonged delivery of cytokines from scaffolds plays an important role for the osteogenesis of stem cell. Bone marrow stem cells (BMSCs) can be differentiated in culture into osteoblasts, chondrocytes, and even myoblasts. In this study, we developed DBP contained poly(L-lactide-co-glycolide) scaffolds for tissue engineered bone. Scaffolds were prepared by solvent casting/salt leaching methods. Characterizations of DBP contained PLGA scaffold were analyed by SEM, mercury porosimeter, and so on. Cellular viability and proliferation were assayed by MTT test. For osteogenic differentiation, BMSCs were cultured with osteogenic medium included DMEM, 10% FBS, 10-8M dexamethasone, 10-8M vitamin D3, 10mM β-glycerophospahte, and 50㎍/㎖ L-ascorbic acid and were analyed by alkaline phosphate activity, histochemical staining, and RT-PCR.
