Hydrocortisone acetate(HCA), an anti-inflammatory agent degradable in water, was tested for its stability in the liposomal system. Three commonly applied dermatological semi-solid bases, such as Hydrogel, Cream, and Poly(ethylene glycol)(PEG) hydrophilic ointment, were prepared and mixed with liposomal suspensions containing HCA. The three samples were stored at room temperature, 30 deg.C, and 40 deg.C for 4 weeks. The stability of the drug as well as of the liposomes in the bases was tested with HPLC, and the effects of liposome entrapment, types of bases, base viscosity, and lipid purity on the drug stability were investigated. The HPLC analysis results for the HCA concentrations in those three samples showed that HCA is stable as being entrapped in liposomes. The protective hydrophobic lipid bilayer seem to have prevented the contact between HCA and the aqueous phase. The liposomal stability was shown to be the best in hydrogel among the three base samples.
