Magnetic separation using functionalized magnetic bead is an efficient technology for capturing specific biomolecules in a complex mixture. However, non-specific binding of proteins has often been a serious problem in the bio-separation process. In this study, we attempted to find suitable surface modification methods for the protein-resistant magnetic beads. Firstly, monodisperse poly(styrene-divinylbenzene) beads (PS-DVB) were prepared by seeded emulsion polymerization (6.5-7.0 μm). The PS-DVB beads were sulfonated and subsequently magnetized using Fe2+/Fe3+ aqueous solution. Thereafter, poly(ethyleneglycol) (PEG) group was introduced onto the magnetized beads by either coupling of diamino-PEG to the sulfonated group or surface-grafting using PEG macromers. We examined if the beads might have the property of reducing the non-specific adsorption of protein. |
