Glycosylceramides, such as glucosylceramide, galactosylceramie, GM1 and etc, are known to have many beneficial biological effects on human body. For instance, effects of galactosylceramide include immunomodulatory effects, natural T killer cells activation and increasing antibody productions. GM1, which has the form of oligosaccharides attached to ceramide, has effects on neurodegenerative condition, parkinson’s disease, substantia nigra degeneration and cellular damage. The aim of this study is to set up an enzymatic synthesis to produce galactosylceramide in bacteria. The author is currently screening for ceramide N deacylase/ceramidase that have the function to synthesize ceramide with the reverse hydrolysis activity. Ceramides synthesized by those deacylase/ceramidase are the acceptor substrates in the production of galactosylceramide. Ceramide galactosyltransferase or galactosylceramide synthase could then transfer galactose to ceramide for the production of the desired target. This enzymatic synthesis system of galactosylceramide could be the potential platform in producing other various forms of glycosylceramides. |
