Immobilization of Candida rugosa ipase by Ca-alginate |

Lipase (E.C. 3.1.1.3) from Candida rugosa was entrapped in Ca-alginate beads. Alginateis a naturally derived linear polysaccharide composed of (1-4)-linked β-D-mannuronic acid and α-L-guluronic acid. It has been extensively used as biomaterial support due to their mild gelling properties and nontoxicity. Ca-alginate beads were prepared by ionic gelation using calcium chloride. The activities of native and immobilized enzyme were determined by catalyzing olive oil via hydrolysis in an aqueous medium and the immobilization efficiency was measured by BCA assay. Effects of CaCl2 concentration, Na-alginate content, ratio of enzyme to Na-alginate, and bead size were investigated in this study. The activity of lipase-alginate beads increased with decreasing Ca2+ concentration and bead size. |
