제목 A Controlled Pulsatile Pressure Bioreactor for Tissue-engineered Vascular Grafts using Cell matrix Engineering
초록 To overcome limitation associated to the nano-structrue scaffold in this study, we adapted two concepts which were matrix sheet engineering and dynamic culture. After SMCs seeded onto the 2D PLCL electrospun scaffold, it rolled to have 3D shape as like a native vessel. We developed controlled pulsatile bioreactor that produced static flow and dynamic flow. Electrospun scaffolds were seeded with SMCs, cultured under dynamic conditions or static conditions for 14 days and evaluated. Mechanical examination showed that vascular grafts under dynamic culture groups had higher burst strength and E-modulus than static groups. Also histological examination showed that the rate of proliferation of SMCs under dynamic condition was better than static condition. We confirmed that new designed pump produced pulsatile pressure similar to physiological condition and scaffold constructed 3D shaped vascular grafts could be useful in vascular tissue engineering.
