제목 Hyaluronic acid (HA) hydrogel with electrospun PCL fibers platform for understanding brain tumor microenvironment
초록 One of heterogeneous subpopulation of malignant brain tumor, the glioma cancer stem cells (GSCs), have been reported to be responsible for therapeutic resistance. However, the microenvironmental effects on GSCs still remain unknown due to a lack of plausible in vitro models. To investigate these features of GSCs, we thus propose 3D in vitro platform, which mimic the brain tumor microenvironment by incorporating the major proinvasive cues: HA-rich ECM and white matter tract. GSCs invaded aggressively within HA-rich hydrogel, comparing to non-HA hydrogel. When GSCs were cultured within PCL fibers-incorporated HA-rich hydrogel, which mimic the topography of white matter tract, the F-actin of GSCs were highly aligned and guided along the fibers. With the proposed platform, we observed interactions of GSCs with their in vivo-like microenvironments, ultimately contributing to find a proper anti-cancer treatment to patients.
