Drug-inorganic layered hydroxide hybrid in cancer chemotherapy: a novel strategy to suppress drug resistance |

Inorganic layered double hydroxides (LDHs) are biocompatible layered materials which have attracted increasing attention as drug delivery nanovehicles. In this study, we evaluated the methotrexate (MTX)-LDH hybrid system in terms of its effects on cellular uptake, retention, and the efficacy of MTX, a well known anticancer drug, in human osteosarcoma cell lines which are MTX-sensitive and even MTX-resistant and explored the underlying mechanism for the effects. Our studies demonstrated that the efficacy of MTX against both sensitive and resistant cell lines increased significantly in MTX-LDH hybrids compared to an equivalent free MTX. Moreover, MTX-LDH hybrids considerably enhanced the cellular uptake of MTX, and well retained in both sensitive and resistant cell lines than the free MTX. The present study suggests a new mechanism for the suppression of drug resistance in tumor cells by using inorganic nanoparticle/anti-cancer drug hybrid system. |
