제목 Stent containing CD44-targeted polymeric prodrug releasing paclitaxel and gemcitabine in a time interval controlled manner for synergistic human biliary cancer therapy
초록 Drug eluting stents (DES) have been suggested as promising strategy for human biliary cancer. However, the implementation of DES has two major problems, side effects of drugs and difficulty to control drug release. In this report, we designed temporally controlled polymeric multi prodrug nanoparticle (TCMPN) eluting stents consisting of polyurethane (PETE) nano-fiber as a polymeric matrix and paclitaxel (PTX) loaded CD44-targeting hyaluronic acid conjugated poly (lactic-co-glycolic acid) and gemcitabine (PLGA-HA-GEM). TCMPN enabled temporal release, that PTX was released first due to the collapse of structure in endosomes and GEM was hydrolyzed from PLGA-HA-GEM later in response to low pH, inducing synergistic anticancer activities and when being embedded in PETE nano-fiber showed sustainable prodrug release. These results indicate that this technology will open up avenues for cancer therapy by combining localized delivery and functional multi drug loaded nanoparticles.
